Evaluation of a bone filler scaffold for local antibiotic delivery to prevent Staphylococcus aureus infection in a contaminated bone defect

Abstract We previously reported the development of an osteogenic bone filler scaffold consisting degradable polyurethane, hydroxyapatite, and decellularized bovine particles. The current study was aimed at evaluating use this as a means local antibiotic delivery to prevent infection in defect contaminated with Staphylococcus aureus . evaluated two formulations same component ratios but differing overall porosity surface area. Studies vancomycin, daptomycin, gentamicin confirmed that uptake concentration dependent increased correlated prolonged release. also demonstrate vancomycin can be passively loaded into either formulation sufficient rabbit model segmental defect. Moreover, even those few cases which complete eradication not achieved, number viable bacteria significantly reduced by treatment there no radiographic evidence osteomyelitis. Radiographs microcomputed tomography (µCT) analysis from vivo studies suggested addition did have any significant effect on itself. These results potential utility our regeneration for defects.
